General description

Mast cells play an important role in innate immunity or the response of the body to invading pathogens. The functions of mast cells are regulated by a number of surface receptors; c-Kit (Stem cell factor receptor, SCF R; CD117) being the most critical. The signalling triggered by c-Kit results in mast cell activation in response to antigens. The response involves tyrosine phosphorylation by specific tyrosine kinases that drive the downstream signalling process. Tyrosine 823 is a major autophosphorylation site of c-Kit. The secondary pathways activated by c-Kit include Akt, mTOR, MAPK, Sos-Grb2 and c-Jun. In addition to development of mast cells, c-Kit also regulates their subsequent maturation and homing to the residence sites. Constitutive activation by SCF R is implicated in myeloproliferative discorders, systemic mastocytosis and gastrointestinal stomal cell tumors
The antibody detects human c-Kit (145 kDa). Cow and dog (100% homologous), mouse, rat and chicken (92% homologous) have not been tested, but are expected to react.

Immunogen

synthetic phosphopeptide derived from the region of human c-Kit that contains tyrosine 823.

Application

Anti-phospho-c-Kit (SCFR) pTyr823 antibody may be used for immunoblotting at a working concentration of 0.1-1.0 μg/ml.

Physical form

Solution in Dulbecco′s phosphate buffered saline (without Mg2+ and Ca2+), pH 7.3, with 1.0 mg/mL BSA (IgG and protease free) and 0.05% sodium azide.
