General description

CD30 is a type I transmembrane glycoprotein of the TNF receptor superfamily. The extracellular domain of recombinant mouse CD30/Fc is fused to the carboxyterminal 6X histidine-tagged Fc portion of human IgG1 by a polypeptide linker.

Specificity

Shows no cross-reactivity with recombinant human CD30. The antibody will stimulate the bioactivity of mouse splenic B cell proliferation in the presence of IL-4 and IL-5.

Immunogen

recombinant mouse CD30, extracellular domain, expressed in NSO cells.

Application

Anti-CD30 is suitable for western blotting at a concentration of 0.1-0.2μg/mL and indirect ELISA at a concentration of 0.5-1.0μg/mL

Biochem/physiol Actions

Anti-CD30 recognizes recombinant mouse CD30 by immunoblotting and ELISA. In immunoblotting and ELISA, this antibody does not cross-react with recombinant human CD30. Reduced mouse CD30/Fc monomer has a calculated molecular mass of approximately 55 kDa. Due to glycosylation, the recombinant protein migrates as an approximately 90 -100 kDa protein in SDS-PAGE under reducing conditions. CD30 specific IgG is purified from goat serum using mouse CD30 affinity chromatography. CD30 has a critical role in the pathophysiology of Hodgkin′s disease and other CD30+ lymphomas. CD30 acts as a costimulatory molecule in thymic negative selection. Anti-CD30 is developed in goat using purified recombinant mouse CD30 extracellular domain, expressed in mouse NSO cells as immunogen.

Physical form

Lyophilized from a 0.2 μm filtered solution in phosphate buffered saline

Reconstitution

Reconstitute with 1 mL sterile PBS to yield 0.1 mg/ml solution.

Murine CD30 cDNA predicts a protein of 498 amino acids with homology to the TNF receptor family of proteins characterized by repeated cysteine-rich motifs in the extracellular domain. Murine CD30, although homologous to human CD30, has a 90 amino acid

The biological function of CD30 in the thymus has been only partially elucidated, although recent data indicate that it may be involved in negative selection. Because CD30 is expressed only by a small subpopulation of medullary thymocytes, we generated transgenic
