Application

A40926 is an antibiotic that is an analog of teicoplanin and the precursor to dalbavanain. It is a mixture of related glycopeptides isolated from Nonomuraea sp. A40926 is used to study mechanisms of glycopeptide resistance [1], the activation of antibiotic production [2] and is used in pharmokinetic studies[3].

Biochem/physiol Actions

A40926 is effective against Gram positive bacteria, and was found to be highly bactericidal against Neisseria gonorrhoeae. The mechanism of action is via inhibition of bacterial cell wall formation.
A40926 is effective against Gram positive bacteria, and was found to be highly bactericidal against Neisseria gonorrhoeae.[4] The mechanism of action is via inhibition of bacterial cell wall formation. A40926 is a complex of four main factors. These factors contain a fatty acid as part of a glycolipid attached to the peptide backbone.

In glycopeptide-resistant enterococci and staphylococci, high-level resistance is achieved by replacing the C-terminal d-alanyl-d-alanine of lipid II with d-alanyl-d-lactate, thus reducing glycopeptide affinity for cell wall targets. Reorganization of the cell wall in these organisms is directed by the vanHAX
