General description

LIN-28 is a RNA-binding protein that acts as an inhibitor of let-7 tumor suppressor microRNAs, and posttranscriptional regulator of mRNAs involved in the renewal and development of embryonic stem cells.[1][2][3][4][5] Anti-LIN28 reactivity is seen in extragonadal seminomas/germinomas, embryonal carcinoma, teratomas, and yolk sac tumors. Anti-LIN28 reactivity is only seen in a small fraction of various extragonadal-non-germ cell tumors, and adult testis tissues. Therefore, Anti-LIN28 is a useful marker in the differential identification of germ cell tumors and cancers of non-germ cell origin.[6][7][8][9][10][11][12]

OCT3/4, NANOG, SOX2 and, most recently, LIN28 have been identified as key regulators of pluripotency in mammalian embryonic and induced stem cells, and are proven to be crucial for generation of the mouse germ-cell lineage. These factors are a hallmark

The let-7 tumor suppressor microRNAs are known for their regulation of oncogenes, while the RNA-binding proteins Lin28a/b promote malignancy by inhibiting let-7 biogenesis. We have uncovered unexpected roles for the Lin28/let-7 pathway in regulating metabolism. When overexpressed in mice, both

Lin28 inhibits the expression of let-7 microRNAs but also exhibits let-7-independent functions. Using immunoprecipitation and deep sequencing, we show here that Lin28 preferentially associates with a small subset of cellular mRNAs. Of particular interest are those for ribosomal proteins and
