General description

1 ml settled resin of FabRICATOR (IdeS) immobilized on highly cross-linked agarose and 1 ml CapureSelect IgG-Fc affinity ligand immobilized on agarose. Agarose resins are suspended in 20% ethanol/water (V/V)

Application

FragIT MidiSpin Kit (cleaving and purifying up to 10mg IgG) can be used for antibody characterization.

Biochem/physiol Actions

IgG fragmentation is accompanying with adverse consequences like low yield, time consuming optimization and operation at low pH with loss of immunoreactivity. These consequences can be overcome with FragIT MidiSpin Kit, which comprises of pre-filled Fabricator enzyme covalently coupled to agarose beads that facilitates the cleavage of IgG (cleaving and purifying up to 10mg IgG) to generate pure F(ab′)2 and Fc fragments. FabRICATOR is a digestive enzyme that digests IgG specifically below the hinge region so there is no risk of over digestion if incubation time exceeds.

Unit Definition

1 ml of settled resin fragments ≥ 95% of 5 mg of human IgG in 15 minutes at 25°C, pH 6.6 as monitored by SDS-PAGE.
