The sample solution refers to what is being assayed for inhibition rate, which in this case is serum. To measure the percentage inhibition rate, various concentrations of superoxide dismutase need to be prepared. The superoxide dismutase standard can be prepared using product S7571 at concentrations of 200 U/ml, 100 U/ml, 50 U/ml, 20 U/ml, 10 U/ml, 5 U/ml, 1 U/ml, 0.1 U/ml, 0.05 U/ml, 0.01 U/ml, and 0.001 U/ml.
Sample preparation involves preparing solutions at various concentrations, as an inhibition curve requires multiple points to generate a curve. Samples should be diluted in dilution buffer or saline. Blank 2 is prepared if the sample solution has visible color and should be prepared according to Table 1. For every sample, the corresponding Blank 2 should have the same sample composition. A visual representation of a planned dilution for 1, 1/5, 1/5^2, 1/5^3, 1/5^4, 1/5^5, 1/5^6 can be prepared as a general recommendation.
