Questions

1–2 of 2 Questions
  1. Protocol said that we have to add EGF just before using it for cell culture? Every time we have to add EGF before transferring the medium to keratinocytes. Can't we add it to a 50 ml aliquot and store it at 4 degrees for its use within a week or 10 d?

    1 answer
    1. In order to retain the maximum potency of EGF it is best to store the reconstituted growth factor at 1mg/mL or with a carrier such as BSA at 0.1%, for lower concentrations. Solutions are stable up to 1 week when stored refrigerated. One time use aliquotes may also be stored frozen at -20 deg. C. Avoid repeated freeze/thaw cycles. For an example of product stability for EGF see the link below to an EGF product data sheet:

Please see the product protocol where no supplementation is recommended: https://www.sigmaaldrich.com/technical-documents/protocol/cell-culture-and-cell-culture-analysis/primary-cell-culture/human-epidermal-keratinocytes.
