General description

This proficiency testing sample is produced in accordance with ISO/IEC 17043:2010. All information regarding the use of this material can be found in the reporting packet supplied for each sample.

Preparation Note

The PT sample is provided in duplicate as a stable dehydrated Vitroid requiring hydration. The additional sample is provided for mishaps in the lab. NELAC prohibits the practice of multiple tests on a PT sample unless it is normal laboratory practice and multiple analysis is conducted on normal day to day samples. The Vitroid is the colored disk above the cotton ball. The Vitroid contains the concentrated bacteria and must be hydrated prior to analysis. The opaque beads below the cotton ball are desiccants and should not be analyzed. A 100 g of sterile soil/sediment is supplied with the PT sample. Store the PT sample at 4 °C. The dehydrated Vitroid sample is stable for a time exceeding the study schedule.

Information
Due to the sensitivity of microorganisms and short holding times after hydration, please read entire instructions prior to preparation. This sample is a single Vitroid and the possible concentration range is quite broad, it may be necessary to analyze the sample in a dilution series using general phosphate buffer solution. This sample may be analyzed by any appropriate method for coliforms.

Sample preparation
While the sample is equilibrating to room temperature, warm the dilution water (use general phosphate buffered water) in an incubator to 35-45 °C. This will take about 60 minutes.
Weigh 1 gram of blank soil and add to sterile blender jar, open and add one Vitroid (microorganisms) to jar, and finally add 99 mL sterile phosphate buffer.
Ensure the colored Vitroid dissolves completely.
Blend for 1 to 2 minutes at low speed.
Prepare the appropriate decimal dilutions of the homogenized slurry as quickly as possible to minimize settling.

Other Notes

Proficiency testing samples are presented as single blind. No values for the samples are released until the testing period concludes. Values of analytes vary lot to lot.
