General description

Histone H1 plays an essential role in maintaining and establishing higher order chromatin structures. The linker histone H1 basically binds to the DNA which is entering and exiting the nucleosomal core particle. All H1 variants in higher eukaryotes comprises of a general structure, consisting of a central conserved globular domain and less conserved N-terminal and C-terminal tails. It has been suggested that each H1 variant has a specific function owing to its tail region which is specifically conserved among species, but differ in variants.[1][2]

Application

Lysine-rich preparation used as a substrate for protein kinases.

Histone H1 was used as a substrate in kinase assay.[1][2]
Studies have suggested that histoneH1 might act as a general repressor of transcription.[1][2]

Histone H1 consists of 212 amino acids.
Histone H1 has a very high lysine content (approx. 25%) which contributes to its high basic charge.

Preparation Note

Working concentration: 1 to 5 mg/ml
Working solution: Solvent is recommended in double-distilled water.
Storage conditions (working solution): Aqueous solution of Histone H1 is stable at -15 to -25 °C for at least three months.
Avoid repeated freezing and thawing!
