General description

Tetraspanin-33 (UniProt: Q8R3S2; also known as Tspan-33, Penumbra, mPen, Proerythroblast new membrane) is encoded by the Tspan33 (also known as Pen) gene (Gene ID: 232670) in murine species. Tetraspanins are evolutionarily conserved proteins and thus far 33 of them have been described in human and murine species. Tspan-33 is a multi-pass, disulfide-linked homodimeric membrane protein of the tetraspanin (TM4SF) family. It has been reported that Tspan-33 is highly expressed in activated B cells. Its expression has also been reported in malignant B cells. Two isoforms of Tspan-33 have been described that are produced by alternative splicing. Tspan-33 plays an important role in normal erythropoiesis and in the differentiation of erythroid progenitors. Tspan-33 contains four transmembrane domains (aa 25-45, 65-85, 97-117, 236-256), three cytoplasmic domains (aa 1-24, 86-96, 257-283), and two extracellular domains (aa 46-64, 118-235). Several tetraspanins interact with specific membrane proteins via their second extracellular domain (aa 118-235) and incorporate them into tetraspanin-enriched microdomains to modulate the effectiveness of signal transduction by those membrane proteins. Specificity

Clone 29A6.2 specifically detects Tspan-33 and targets an epitope with in the extracellular domain.

BaF3 cells expressing full-length murine Tspan-33 fused with a Myc-His tag at the C-terminus.
