General description

Transmembrane protein 64 (UniProt Q6YI46) is encoded by the TMEM64 gene (Gene ID 169200) in human. TMEM64 is a 6-transmembrane protein that plays a role in modulating Wnt/ -catenin signaling-mediated osteoblast and adipocyte differentiation. TMEM64 interacts and positively regulates sarcoplasmic endoplasmic reticulum Ca(2+) ATPase 2 (SERCA2) activity, which is important for RANKL-induced Ca(2+) oscillation, Ca(2+)/calmodulin-dependent protein kinases (CaMK) IV activity and mitochondrial ROS formation needed for for osteoclast formation. Mice with Tmem64 deficiency are characterized by a high-bone-mass phenotype, displaying higher levels of ALP, osteocalcin (Ocn), and type 1 collagen (Col 1a) transcripts than in wild-type mice. Tmem64-knockout in mice is reported to markedly enhance osteoblast differentiation and inhibit adipocyte differentiation from bone marrow-derived stromal cells (BMSCs), resulting in a preferential switch of mesenchymal progenitors favoring the osteogenic over the adipogenic lineage.

Specificity

The immunogen sequence is 100% conserved between human and rat species and present in all four human spliced isoforms reported by UniProt (Q6YI46 & D3ZV19). Murine cross-reactivity is highly likely, but has not been confirmed.
