Gene Information

human ... LANCL2(55915)

General description

LanC-like protein 2 (UniProt: Q9NS86; also known as Testis-specific adriamycin sensitivity protein) is encoded by the LANCL2 (also known as GPR69B, TASP) gene (Gene ID: 55915) in human. LANC2 is a member of the highly conserved and widely distributed lanthionine synthetase component C-like (LANCL) protein family. It is well expressed in brain and testis and is localized to the plasma membrane, juxta-nuclear vesicles, and the nucleus. The N-terminus of LANCL2 differs from the other LANCL proteins by an N-terminal extension of 17 amino acid residues and an insertion of 19 residues. LANCL2 contains 15 cysteine residues, three of which are conserved in all human LANCL proteins and 8 are conserved between LANCL2 and LANCL1. LANCL2 displays about 75% amino acid homology with LANCL1 protein. LANCL2 contains a phosphatidylinositol phosphate (PIP) binding site at the N-terminus and its myristoylation is essential for its membrane association. LANCL2 is essential for abscisic acid (ABA) binding on the cell membrane and activation of the ABA signaling pathway in granulocytes. Exogenous expression LANCL2 in a sarcoma cell line is shown to reduce the expression of ABCB1 (P-glycoprotein 1) and increase cellular sensitivity to adriamycin. (Ref.: Sturla, L et al. (2009). J. Biol. Chem. 284(41):28045-57; Ladlinger, C et al. (2006). Biochim Biophys Acta. 1758(11):1759-67).
