General description

Anaphase-promoting complex subunit 4 (UniProt Q9UJX5; also known as APC4, Cyclosome subunit 4) is encoded by the ANAPC4 (also known as APC4) gene (Gene ID 29945) in human. APC4 is a component of the anaphase-promoting complex or cyclosome (APC/C). APC/C is an E3 ubiquitin ligase complex that promotes the transition from metaphase to anaphase of the cell cycle by targeting specific proteins for proteasomal degradation at specific times under the control of the spindle assembly checkpoint (SAC). The catalytic core of the APC/C consists of the cullin subunit APC2 and RING H2 domain subunit APC11. Other APC/C components, including APC4, are composed multiple repeat motifs with the main purpose of providing molecular scaffold support. APC4 contains WD repeats and has a molecular weight of approximately 92 kDa. APC4 is located in the nucleus during interphase and at the centrosome during metaphase/anaphase.

Specificity

Clone CIV1.1 target human APC4 C-terminal end sequence present in spliced isoforms 1 and 3, but absent in isoform 2 reported by UniProt (Q9UJX5).

Linear peptide corresponding to human APC4 C-terminal end sequence.

Application

Anti-APC4, clone CIV1.1 Antibody, Cat. No. MABS1257, is a highly specific mouse monoclonal antibody that targets APC4 and has been tested in Immunohistochemistry (Paraffin), Immunoprecipitation, and Western Blotting.
Immunohistochemistry Analysis: A 1:250 dilution from a representative lot detected APC4 in human spleen tissue section.

Immunoprecipitation Analysis: A representative lot co-immunoprecipitated Cdc20, Mad2, and BubR1 with APC4 from prometaphase HeLa cells (Di Fiore, B., et al. (2015). Dev. Cell. 32(3):358-372).

Western Blotting Analysis: A representative lot detected APC4 in whole cell lysates, as well as in FLAG-tagged cyclin A and FLAG-tagged BubR1 immunoprecipitates from prometaphase HeLa cells (Di Fiore, B., et al. (2015). Dev. Cell. 32(3):358-372).

Quality

Evaluated by Immunohistochemistry in human uterus tissue section.

Immunohistochemistry Analysis: A 1:250 dilution of this antibody detected APC4 in human uterus tissue section.
