General description

Protein phosphatase 1D (EC 3.1.3.16; UniProt O15297; also known as Wild-type p53-induced protein phosphatase 1, PP2C-delta, Protein phosphatase 2C isoform delta, Protein phosphatase magnesium-dependent 1 delta) is encoded by the PPM1D (also known as WIP1) gene (Gene ID 8493) in human. Wild-type p53-induced phosphatase 1 (WIP1) or PP2C-delta is a serine/threonine phosphatase. Unlike PP2A or PP1 enzymes, WIP1 functions as a monomer whose catalytic activity is dependents on Mg2+ or Mn2+ and is insensitive to inhibition by okadaic acid or microcystin. Casein kinase 2 (CK2) andPP2C-delta catalyze the phosphorylation and dephosphorylation, respectively, of LSD1 on Ser131 and Ser137 residues. S131/S137 phosphorylation promots LSD1 interaction with RNF168 and RNF168-dependent 53BP1 ubiquitination and subsequent recruitment to DNA damage sites. Other known PP2C-delta targets include ATM (S367, S1981), -H2AX, CHK1 (S345) and CHK2 (T68).

Specificity

Clone 2F7.1 targets an internal epitope conserved among the two human spliced isoforms and mouse/rat PP2C-delta (Uniprot O15297, B1WCA0, Q9QZ67).

GST-tagged recombinant human PP2C-delta internal fragment.

Application

Anti-PP2C Isoform delta Antibody, clone 2F7.1, Cat. No. MABS1189, is a highly specific mouse monoclonal antibody, that targets PPM1D and has been tested in Western Blotting and Immunohistochemistry (Paraffin).
Immunohistochemistry Analysis: A 1:1,000 dilution from a representative lot detected PP2C-delta in human prostate cancer, normal human brain and rat testis tissue sections.

Evaluated by Western Blotting in HL-60 human promyelocytic leukemia cell lysate.

Western Blotting Analysis: A 1:500 dilution of this antibody detected PP2C-delta in 10 µg of HL-60 human promyelocytic leukemia cell lysate.

~60 kDa observed. 66.68/47.21 kDa (human isoform 1/2), 65.72 kDa (mouse), and 65.68 kDa (rat) calculated.
