General description

Sulfonylurea receptor 2 (Sur2), also known as ATP-binding cassette sub-family C member 9 (ABCC9), belongs to the ATP-binding cassette transporter (ABC transporter) superfamily. Sur2 is a subunit of ATP-sensitive potassium channels (KATP). The potassium channel, inwardly rectifying subfamily J member 11 (KCNJ11) interacts with Sur2 in the formation of cardiac and smooth muscle-type KATP channels, where Sur2 is required for activation and regulation of the channel. Sur2 is expressed at high levels in heart, skeletal muscle and ovary, and at moderate levels in brain, tongue and pancreatic islets. Expression in other tissue is at low levels. Defects in KCNJ11 are the cause of familial hyperinsulinemic hypoglycemia type 2 (HHF2), diabetes mellitus permanent neonatal (PNDM), as well as transient neonatal diabetes mellitus type 3 (TNDM3). Sur2B is an alternatively spliced isoforms of Sur2 (Sur2A), having differences in the amino acid sequence (1504-1545).

Specificity

This antibody recognizes the cytoplasmic domain of the cleaved form of SUR2B. This antibody may cross react with SUR1 based upon 75% sequence homology.

Immunogen

Recombinant protein corresponding to the cytoplasmic domain of the cleaved form of rat SUR2.

Application

Anti-SUR2B Antibody, clone N323A/31 is a highly specific mouse monoclonal antibody, that targets SUR & has been tested in western blotting & IHC.
Western Blotting Analysis: 1 µg/mL from a representative lot detected the cleaved form of SUR2B in mouse brain tissue lysate.

Immunohistochemistry Analysis: A 1:1,000 dilution from a representative lot detected the cleaved form of SUR2B in rat skeletal muscle tissue.

Western Blotting Analysis: A representative lot detected SUR2B in wild type mouse heart tissue lysate, and demonstrated a loss of signal in SUR2B knock out mouse heart tissue lysate (Prof. J. Trimmer, University of California, Davis.).

Quality

Evaluated by Western Blotting in human kidney tissue lysate.

Western Blotting Analysis: 1 µg/mL of this antibody detected the cleaved form of SUR2B in 10 µg of human kidney tissue lysate.

Target description

~58 kDa observed. The calculated molecular weight is 175 kDa However, this protein may be observed at 58 kDa due to proteolytic cleavage (Prof. J. Trimmer, University of California, Davis.).
