Description

General description

Neuron navigator 1 (UniProt Q8NEY1; also known as Pore membrane and/or filament-interacting-like protein 3, Steerin-1, Unc-53 homolog 1, unc53H1) is encoded by the NAV1 (also known as KIAA1151, KIAA1213, POMFIL3, STEERIN1, UNC53H1) gene (Gene ID 89796) in human. NAV1 is a member of the family of proteins called navigators that play a role in mediating neurite outgrowth. NAV1 is shown to interact with the guanine nucleotide exchange factor (GEF) TRIO, whereby it enhances the affinity of TRIO for Rho family GTPases Rac1 and RhoG. The plus-end-tracking protein (+TIP) EB1 at the ends of growing microtubules recruits TRIO, facilitating the formation and localization of functional NAV1-TRIO complexes, which in turn regulates neurite outgrowth by selectively activating Rac1 at the growth cone.

Specificity

Expected to react with all seven spliced isoforms of murine NAV1 (UniProt Q8NEY1-1 through Q8NEY1-7), but not NAV2 or NAV3.

Immunogen

His-tagged recombinant protein corresponding to the Internal region of human NAV1.

Application

Anti-NAV1 Antibody, clone 7F12 (KT124) is an antibody against NAV1 for use in Immunocytochemistry, Western Blotting.

Western Blotting Analysis: 1.0 µg/mL from a representative lot detected NAV1 in 10 µg of Murine NIE-115 neuroblastoma cell lysate.
Immunocytochemistry Analysis: 10.0 µg/mL from a representative lot detected NAV1 in Neuro-2a and NIE-115 cells.
Western Blotting Analysis: A representative lot detected endogenous NAV1 protein in murine neuroblastoma N1E-115 cell lysate, as well as exogenously expressed NAV1, but not NAV2 or NAV3, GFP fusion protein in lysates from transfected HEK293T cells (van Haren, J., et al. (2014). Curr. Biol. 24(15):1778-1785).
Immunocytochemistry Analysis: A representative lot detected NAV1 accumulation in cellular extensions of primary murine hippocampal neurons and murine neuroblastoma N1E-115 cells (van Haren, J., et al. (2014). Curr. Biol. 24(15):1778-1785).

Quality

Evaluated by Western Blotting in murine Neuro-2a (N2a) neuroblastoma cell lysate.

Western Blotting Analysis: 1.0 µg/mL of this antibody detected NAV1 in 10 µg of murine Neuro-2a (N2a) neuroblastoma cell lysate.

Target description

~240/150 kDa observed. 202.5 kDa (isoform 1), 201.5 kDa (isoform 2), 202.1 kDa (isoform 3), 196.0 kDa (isoform 4), 160.8 kDa (isoform 5), 126.2 kDa (isoform 6), 195.0 kDa (isoform 7) calculated.
