General description

Cdc42-interacting protein 4 (UniProt Q15642; also known as hSTP, CIP4, Protein Felic, Salt tolerant protein, Thyroid receptor-interacting protein 10, TR-interacting protein 10, TRIP-10) is encoded by the TRIP10 (also known as CIP4, STOT, STP) gene (Gene ID 9322) in human. Cdc42-interacting protein 4 (CIP4) is a member of the F-BAR family of proteins. These proteins contain a hydrophobic N-terminal, alpha-helical region that is related to the Bin Amphiphysin Rvs (BAR) protein family. The F-BAR domain binds the phospholipid membrane with its concave surface. CIP4 contains an SH3 domain that has been reported to play a role in endocytosis; an REM (Hr1) repeat, and an FCH domain, which facilitates in microtubule binding. CIP4 plays several other important roles in cells. It is an essential component of insulin signaling for GLUT4 translocation to the plasma membrane and is also required during endocytosis for the orchestration of membrane tabulation with the remodeling of the actin cytoskeleton. It is shown to binds to lipids such as phosphatidylinositol 4, 5-bisphosphate and phosphatidylserine and promotes membrane invagination and the formation of tubules.

Specificity

Clone 8F10.1 targeted region is present in all five human and all four murine spliced isoforms reported by UniProt (Q15642 and Q8CJ53).

Immunogen

GST-tagged recombinant human CIP4 fragment corresponding to a region within the F-BAR domain.

Application

Anti-CIP4, clone 8F10.1, Cat. No. MABN1555, is a highly specific mouse monoclonal antibody that targets CIP4 and has been tested in Immunohistochemistry (Paraffin) and Western Blotting.
Immunohistochemistry Analysis: A 1:250 dilution from a representative lot detected CIP4 in human pancreas, small intestine, and tonsil tissue sections.

Western Blotting Analysis: 2 µg/mL from a representative lot detected multiple CIP4 isoforms in 10 µg of Neuro-2a (N2a) mouse neuroblastoma cell lysate.

Quality

Evaluated by Western Blotting in Neuro-2a mouse neuroblastoma cell lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected CIP4 in 10 µg of Neuro-2a (N2a) mouse neuroblastoma cell lysate.

Target description

~75/70/50/38 kDa observed. 68.35/62.59/67.60/52.70/39.58 kDa (human isoform 1/2/3/4/5), 68.49/68.40/62.65/62.57 kDa (mouse isoform 1/2/3/4) calculated. Uncharacterized bands may be observed in some lysate(s).
