General description

RNA polymerase II (RNAPII or Pol II) is a multi-subunit enzyme responsible for the transcription of transcription of DNA into RNA from protein-coding genes. Transcription initiation requires recruitment of the complete transcription machinery to a promoter via solicitation by activators and chromatin remodeling factors. RNAPII can coordinate 10 to 14 subunits. This complex interacts with the promoter regions of genes and a variety of elements and transcription factors. The DNA binding domain of the polymerase is a groove where TFIIB orients the DNA for unwinding and transcription.

Immunogen

Ovalbumin-conjugated linear peptide corresponding to human RNAPII phosphorylated at Tyr1.

Application

Detect the phospho-RNAPII (Tyr1) protein using this Anti-phospho-RNAPII (Tyr1), clone 3D12 validated for use in WB, IP & ChIP.
Immunoprecipitation Analysis: A representative lot from an independent laboratory detected phospho-RNAPII (Tyr1) in proliferating yeast whole cell extract (Mayer, A., et al. (2012). Science. 336(6089):1723-1725.).

Chromatin Immunoprecipitation (ChIP) Analysis: A representative lot from an independent laboratory immunoprecipitated phospho-RNAPII (Tyr1) in proliferating yeast whole cell extract (Mayer, A., et al. (2012). Science. 336(6089):1723-1725.).

Quality

Evaluated by Western Blot Analysis in untreated and lambda phosphatase treated HeLa cell lysates.

Western Blot Analysis: 2 µg/mL of this antibody detected phospho-RNAPII (Tyr1) in 10 µg of untreated HeLa cell lysate. (Lambda phosphatase treatment was performed during the western blot procedure.)
