General description

Ubiquitin carboxyl-terminal hydrolase BAP1 (EC 3.4.19.12; UniProt Q92560; also known as BRCA1-associated protein 1, Cerebral protein 6) is encoded by the BAP1 (also known as KIAA0272, hucep-6) gene (Gene ID 8314) in human. Originally identified and named as a BRCA1-interacting protein, BAP1 is a tumor suppressor that participates in DNA-damage response, DNA synthesis and cell cycle progression. BAP1 belongs to the ubiquitin C-terminal hydrolase (UCH) family of deubiquitin enzymes (DUBs), all members of which, including UCH-L1, UCH-L3 and UCH-L5, are characterized by a conserved UCH catalytic domain (CD; BAP1 a.a. 1-235). BAP1 and UCH-L5 also share a conserved C-terminal ULD domain (a.a. 640-710 of BAP1). The region between BAP1 CD and ULD is important for binding to the HCF-1 protein. In addition, BAP1 contains a C-terminal extension (CTE; a.a. 711-729) following its ULD. Study shows that while CTE tethers BAP1 to the nucleosome core, additional interaction with ASXL1 DEUBiquitinase ADaptor domain (DEUBAD) is needed to allow BAP1 substrate processing by increasing it′s affinity towards ubiquitin. In a similar fashion, UCH-L5 activity is regulated via interaction with DEUBAD of RPN13 and INO80G. BAP1 is implicated in transcription regulation due to its interaction with transcription-related proteins, such as FOXK2, HCF-1, OGT, LSD2, MBD-6, MBD-5, ASXL1 and ASXL2. BAP1 loss leads to a variety of cancers, including metastatic cutaneous and uveal melanoma, pleural mesothelioma, renal cell carcinoma and bladder tumours.
