General description

DIS3-like exonuclease 2 (UniProt Q8IYB7; also known as DIS3 mitotic control homolog-like 2, DIS3L2, Family with sequence similarity 6 member A, hDIS3L2) is encoded by the DIS3L2 (also known as FAM6A, PRLMNS) gene (Gene ID 129563) in human. Cytoplasmic mRNAs are regulated by degradations to ensure a fast and efficient control of the transcriptome at the post‐transcriptional level. The degradation process is initiated by deadenylation, followed by decapping, after which mRNA is accessible to digestion by the Xrn1 exonuclease in the 5′‐3′ direction or by the exosome complex in the 3′‐5′ direction. In yeast, the cytoplasmic exosome consists of nine inactive subunits (core ring structure) plus an active ribonuclease Rrp44/Dis3. Three Dis3 isoforms exist in human, DIS3, DIS3L, and DIS3L2. DIS3 is nuclear, and DIS3L and DIS3L2 are cytoplasmic. In addtion to mRNAs, DIS3L2 is also reported to mediate the degradtion of 3′ uridylated let-7 precursor (pre-let-7) miRNA. Lin28 is a pluripotency factor expressed in stem cells and cancer cells, where it helps maintain the cells in an undifferentiated and proliferative state by preventing miRNA let-7 maturation. Lin28 binds to pre-let-7 and recruits 3′ terminal uridylyl transferase, which in turn marks pre-let-7 via 3′ oligouridylation for degradation by DIS3L2. Cellular DIS3L2 depletion causes mitotic abnormalities due to the deregulation of the expression of mitotic control proteins and DIS3L2 gene mutations are linked to the congenital overgrowth disease Perlman syndrome.

Specificity

Expected to react with isoforms 1-4. Reactivity toward isoform 5 has not been determined.

Immunogen

GST-tagged recombinant protein corresponding to the Internal (N-terminal half) of human DIS3-like Exonuclease 2.

Application

Anti-DIS3-like Exonuclease 2 Antibody, clone 9H2.1 is an antibody against DIS3-like Exonuclease 2 Antibody for use in Western Blotting.

Quality

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected DIS3-like Exonuclease 2 in 10 µg of HepG2 cell lysate.

Target description

~100 kDa observed. 99.3 kDa (Isoform 1), 69.0 kDa (Isoform 2), 67.3 kDa (Isoform 3), 27.5 kDa (Isoform 4), 40.5 kDa (Isoform 5) calculated.
