General description

Nucleolin, also known as C23, is the major nucleolar protein of exponentially growing eukaryotic cells. It is a ubiquitous conserved multi-domain phosphoprotein found in all vertebrate species. Nucleolin contains a specific bipartite nuclear localization signal sequence and three different structural domains. Nucleolin is a multifunctional protein involved in most steps of ribosome biogenesis. Nucleolin has been implicated in chromatin structure, rDNA transcription, rRNA maturation, ribosome assembly, cytokinesis, nucleogenesis, cell proliferation and growth, and
nucleo-cytoplasmic transport. Nucleolin also exhibits autodegradation, DNA and RNA helicase activities, and DNA-dependent ATPase activity. Nucleolin activities may be regulated by proteolysis, methylation, ADPribosylation, and phosphorylation by several kinases, including casein kinase II (CK2), p34cdc2, and protein kinase C-ζ.

Application

This Anti-Nucleolin Antibody, clone 364-5-10-5 is validated for use in Western Blotting and Immunohistochemistry (Paraffin) and Immunocytochemistry and Flow Cytometry for the detection of Nucleolin.
Western Blotting Analysis: 1.0 µg/mL from a representative lot detected Nucleolin in 10 µg of NIH/3T3 cell lysate.

Immunohistochemistry Analysis: A 1:250-1,000 dilution from a representative lot detected Nucleolin in human breast cancer and human kidney tissue.

Immunohistochemistry Analysis: A 1:10 dilution from a representative lot detected Nucleolin in ovarian cancer metastasis-infiltrated human lymph node tissue.

Immunocytochemistry Analysis: A 1:250 dilution from a representative lot detected Nucleolin in A431 and HEK293 cells.

Flow Cytometry Analysis: A representative lot detected Nucleolin in NK-92MI cells gated and ungated for CD56.

Quality

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: 1.0 µg/mL of this antibody detected Nucleolin in 10 µg of HeLa cell lysate.

Target description

~100 kDa calculated. This protein has many residues that are post transitionally modified (phosphorylation) and can run higher than the calculated molecular weight of 76 kDa in some lysates or have bands at both 76 and ~100-110 kDa (Qiu, J., et al. (1999) Virology. 257(2):373–382.)
