General description

Calpains are non-lysosomal cysteine proteeases which absolutely require calcium for activity. There are two major calpain isozymes: μ-calpain and m-calpain, which require micromolar or millimolar calicum for activity, respectively, in invitro assays. The major calpain isozymes are composed of two subunits: an 80kDa catalytic subunit; and a 30kDa regulatory subunit. The large and small subunits remain associated in the presence of calcium after proteolytic activation of the zymogen.

Specificity

Reacts with human calpain-I (μ-Calpain). P9 does not cross with human placental m-calpain (calpain II). This antibody will immunoprecipitate proteolytically active calpains.

Application

Anti-Calpain I Antibody, large subunit, clone P9 is an antibody against Calpain I for use in ELISA, IP, WB & IC.
Immunoblot: 1:1,000 Detects an 80kDa large subunit μ-calpain, and will also detect the subunit in the non-activated proform of the zymogen, showing a band of ~110kDa. WI-38 human fibroblasts, or human erthryocytes are suggested as positive controls. Immunocytochemistry: 1:100 on A431 epidermal carcinoma.

ELISA: 1:6,000 (against the immunogen)

Optimal working dilutions must be determined by the end user.
