General description

Methylated-DNA-protein-cysteine methyltransferase (EC 2.1.1.63; UniProt P16455; also known as 6-O-methylguanine-DNA methyltransferase, GATase, MGMT, O-6-methylguanine-DNA-alkyltransferase) is encoded by the MGMT gene (Gene ID 4255) in human. MGMT is a DNA repair enzyme that removes alkyl groups from O6-guanine and plays a fundamental role in maintaining genomic integrity. MGMT converts the naturally occurring mutagenic DNA lesion O6-methylguanine back to guanine, thereby prevents mismatch and errors during DNA replication and transcription. MGMT expression level correlates with cellular resistance to alkylnitrosoureas, compounds that induce O6-alkylguanine lesions. MGMT transcription is negatively regulated via promoter methylation, MGMT promoter methylation level is reported to correclate with the survival rate of glioblastoma patients receiving treatment with alkylating agents.

Specificity

Clone MT 3.1 detected p53 expression-dependent MGMT downregulation in IMR90 cells (Harris, L.C., et al. (1996). Cancer Res. 56(9):2029-2032).

GATase purified from CEM-CCRF human leukemic lymphoblasts (Harris, L.C., et al. (1996). Cancer Res. 56(9):2029-2032; Brent, T.P., et al. (1990). Cancer Res. 50(1):58-61).

Application

Anti-MGMT Antibody, clone MT 3.1, Alexa Fluor 647 Conjugate, Cat. No. MAB16200-AF647, is a highly specific mouse monoclonal antibody that targets MGMT and has been tested in Flow Cytometry and Immunocytochemistry.
Flow Cytometry Analysis: 0.2 µL from a representative lot detected MGMT in one million MCF-7 cells.

This monoclonal antibody is also available in unconjugated and Alexa Fluor 488-conjugated forms (Cat. No. MAB16200 and MAB16200-AF488) for flow cytometry, immunocytochemistry, immunohistochemistry, immunoprecipitation, and Western blotting applications.
