Gene Information

General description

Amelogenin, X isoform/AMELX, also known as Leucine-rich Amelogenin peptide, or LRAP, and encoded by the gene AMELX/AMG/AMGX, is an important protein in the biomineralization of teeth. AMELX appears to regulate the formation of crystallites during the secretory stage of tooth enamel development. AMELX is a secreted extracellular matrix protein that has many isoforms. Interestingly mutations in the AMELX protein are associated with changes in enamel microstructure that are linked with severe dental disease, and the various isoforms of AMELX protein all aid in increasing enamel thickness of the developing tooth.

Immunogen

Recombinant protein corresponding to mouse Amelogenin, X isoform/AMELX.

Application

Anti-Amelogenin, X isoform/AMELX Antibody is an antibody against Amelogenin for use in western blotting & IHC.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Amelogenin, X isoform/AMELX using recombinant mouse Amelogenin179.

Western Blotting Analysis: A representative lot from an independent laboratory detected Amelogenin, X isoform/AMELX in DESC spheres and cervical loop lysates (Chang, J. Y., et al. (2013). J Cell Biol. 288(40):28952-289561.).

Immunohistochemistry Analysis: A representative lot from an independent laboratory detected Amelogenin, X isoform/AMELX in normal ameloblasts in the wild type and the pseudostratified layer and amorphous matrix of the null mice (Rios, H., et al. (2005). Mol Cell. Biol. 25(24):11131-11144.).

Quality

Evaluated by Western Blotting in pig enamel acid extract.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Amelogenin, X isoform/AMELX in pig enamel acid extract.

Target description

~23 kDa observed. At least 3 isoforms may be observed between 19-23 kDa in pig enamel acid extracts (Chang, J. Y., et al. (2013). J Cell Biol. 288(40):28952-289561.). Uncharacterized band(s) may be observed in some cell lysates.

Physical form

Rabbit polyclonal serum with 0.05% sodium azide.
Unpurified

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Tooth loss is a significant health issue currently affecting millions of people worldwide. Artificial dental implants, the current gold standard tooth replacement therapy, do not exhibit many properties of natural teeth and can be associated with complications leading to implant
