General description
Ankyrin-2 (UniProt: Q01484; also known as ANK-2, Ankyrin-B, Brain Ankyrin, Non-erythroid Ankyrin) is encoded by the ANK2 gene (Gene ID: 287) in human. Most ankyrins are typically composed of three structural domains: an amino-terminal domain containing multiple ankyrin repeats; a central region with a highly conserved spectrin binding domain; and a carboxy-terminal regulatory domain, which is the least conserved and subject to variation. ANK-2 is a cytoskeletal protein that is present in plasma membrane of neurons as well as glial cells throughout the brain and it is expressed in fetal brain and in temporal cortex of adult brain. Its expression is also shown in the inner segments of rod photoreceptors in retina and at the apical membrane of airway lung epithelial cells. Four isoforms of ANK-2 have been reported that are produced by alternative splicing. ANK-2 can be phosphorylated at multiple sites by different protein kinases and each phosphorylation event regulates its structure and function. In skeletal muscle, ANK-2 is required for proper localization of DMD and DCTN4 and for the formation and/or stability of a special subset of microtubules associated with costameres and neuromuscular junctions. Mutations in ANK2 gene are known to cause Long QT syndrome 4 (LQT4), which is characterized by a prolonged QT interval on the ECG and polymorphic ventricular arrhythmias. It can cause syncope (temporary loss of consciousness caused by a fall in blood pressure) and sudden death in response to exercise or emotional stress.
