General description

Tether containing UBX domain for GLUT4 (UniProt: Q8VBT9; also known as Alveolar soft part sarcoma chromosomal region candidate gene 1 protein homolog, TUG) is encoded by the Aspscr1 (also known as Tug) gene (Gene ID: 68938) in murine species. TUG is a 60-kDa protein present in the cytosol and on membranes. In the absence of insulin, it binds and sequesters GLUT4 intracellularly. Insulin stimulates the release of GLUT4 from TUG and facilitates GLUT4 translocation to cell membrane. TUG undergoes site-specific endoproteolytic cleavage, which separates a GLUT4-binding, N-terminal region of TUG from a C-terminal region. It has been shown that in 3T3-L1 cells, depletion of TUG can mobilize ~60-nm vesicles to the cell surface and these vesicles carry GLUT4 storage vesicle (GSV) cargoes and fuse with the plasma membrane. GSVs are distinct from other exocytic vesicles. It is shown that during exposure to insulin, GSV cargoes recycle in ~150-nm vesicles characteristic of endosomes and can bypass a TUG-regulated membrane trafficking step. Hence, GLUT4 can participate in two different exocytic pathways, which are active at distinct times after insulin addition in 3T3-L1 cells. It has been shown that TUG can link GLUT4 to Golgi matrix proteins, PIST and Golgin-160. Cleavage of TUG separates an N-terminal region that binds GLUT4 from a C-terminal region that binds Golgin-160 and this cleavage thus untethers GSVs from the Golgi matrix. (Ref.: Bogan, JS et al. Specificity

This polyclonal antibody detects TUG in NIH3T3/L1 cells. It targets an epitope within 29 amino acids from the N-terminal region.

Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
