General description

Iroquois-class homeodomain protein IRX-5 (UniProt Q9JKQ4; also known as Homeodomain protein IRXB2, Iroquois homeobox protein 5) is encoded by the Irx5 (also known as Irxb2) gene (Gene ID 54352) in murine species. Iroquois homeobox (Irx) genes encode a family of homeodomain transcription factors that are conserved from worms to vertebrates. Six IRX genes are located in two clusters in human (IRXA and IRXB) and mouse (IrxA and IrxB) genomes. Irx3 and Irx5 of the IrxB cluster are expressed in the developing mouse limb bud and are essential for proximal and anterior skeletal formation. Irx3 and Irx5 are required to establish early anteroposterior (AP) polarity and specify the anterior progenitor population. Individual Irx genes are also important for certain postnatal cardiac functions in the developing mouse heart, including cardiac repolarization (Irx5) and rapid ventricular conduction (Irx3). Postnatal deletion of Irx3 and Irx5 in the myocardium leads to prolongation of atrioventricular conduction, due in part to activation of expression of the Na(+) channel protein Nav1.5. However, combined postnatal loss of Irx3 and Irx5 results in a restoration of the repolarization gradient that is altered in Irx5 mutant hearts, suggesting that postnatal Irx3 activity can be repressed by Irx5 in mice.

Specificity

This rabbit polyclonal antiserum targets an internal region within the C-terminal half of murine IRX-5/IRXB2.

His-tagged recombinant murine IRX-5/IRXB2 C-terminal fragment GST fusion protein (Chen, C.W., et al. (2005). Dev. Biol. 287(1):48-60).

This rabbit polyclonal Anti-IRX-5 Antibody, Cat. No. ABE1932 detects levels of IRX-5, and has been published and validated for use in Western Blotting.
Western Blotting Analysis: A representative lot was affinity purified and detected an increased IRX-5/IRXB2 expression in postnatal P10 heart tissue lysates from miR-1-2-knockout mice (Zhao, Y., et al. (2007). Cell. 129(2):303-317).

Quality

Evaluated by Western Blotting in postnatal mouse heart tissue lysate.

Western Blotting Analysis: A 1:500 dilution of this antiserum detected IRX-5/IRXB2 in 10 µg of postnatal P10 mouse heart tissue lysate.

~54 kDa observed. 50.76 kDa calculated. Uncharacterized bands may be observed in some lysate(s).

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
