General description

FUN14 domain-containing protein 1 (FUNDC1) is an important autophagy partner for mitophagy, the process whereby mitochondria get recycled after damage. FUNDC1 is localized to the outer mitochondrial membrane and widely expressed in all cells and appears to act as an activator of hypoxia-induced mitophagy. FUNDC1 interacts directly with important autophagy markers such as MAP1LC3B and GABARAP. Phosphorylation at Tyr-18 inactivates FUNDC1 mitophagy activation ability, and it is this dephosphorylation following hypoxia that initiates the interaction with MAP1LC3 family proteins and mitophagy. Recent research shows that FUNDC1 expression is controlled via the microRNA miRNA-137, and that this microRNA is critical for mitophagy and mitochondrial recycling.

This Anti-FUNDC1 Antibody is validated for use in Western Blotting and Immunocytochemistry and Immunoprecipitation for the detection of FUNDC1.
Western Blotting Analysis: A representatitive lot of this antibody was used to detect FUNDC1 in A431, C2C12, HEK293, HeLa, HepG2, L6, Mouse brain, NIH/3T3, PC12, PC3, and RAW264.7 cell lysates. This antibody did not detect FUNDC1 in Human placenta, HUVEC, or Jurkat cell lysates.
Western Blotting Analysis: A representatitive lot of this antibody was used to detect puried FUNDC1 protein from bacteria or HeLa cell lysates (Liu, L., et al. (2012) Nature Cell Biology. 14(2):177-185).
Immunocytochemistry Analysis: A representatitive lot of this antibody was used to detect FUNDC1 in HeLa cells (Liu, L., et al. (2012) Nature Cell Biology. 14(2):177-185.
Immunoprecipitation Analysis: A representatitive lot of this antibody was used to co-immunoprecipitate FUNDC1 with endogenous LC3 in HeLa cells that were exposed to normoxic or hypoxic conditions for 24 hrs (Liu, L., et al. (2012) Nature Cell Biology. 14(2):177-185).

Quality

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis: A 1:1000 dilution of this antibody detected FUNDC1 in 10 µg of HepG2 cell lysate.

Target description

~17 kDa observed.
Uncharacterized bands may be visitible in some lysates.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
