General description

The protein named HPGD, or also known as 15-PGDH, or Prostaglandin dehydrogenase 1, and encoded by the gene HPGD/PGDH1, is a protein necessary for the regulation of prostaglandin levels. Prostaglandins are potent inflammatory agents, thus their regulation is key for control inflammatory responses. HPGD works to mediate prostaglandin inactivation by catalyzing the NAD dependent dehydrogenation of lipoxin A4 to form 15-oxo-lipoin A4. Additionally, HPGD inhibits in vivo proliferation of colon cancer cells. HPGD is localized to the cytoplasm. HPGD is expressed in intestinal and colon tissues. HPGD is down regulated in colon cancer and thus is being examined as a therapeutic agent. Defects in HPGD may be associated with Hypertrophic osteoarthropathy, a disease characterized by digital clubbing, periostosis, acroosteolysis, painful joint enlargement, and variable features of pachydermia that include thickened facial skin and a thickened scalp. Other developmental anomalies include delayed closure of the cranial sutures and congenital heart disease. Other diseases associated with this gene’s defects are Crainoosteoarthropathy (swollen joint disease) and congenital nail clubbing where one has very large nails on the fingers and toes.
