General description

Mothers against decapentaplegic homolog 2 (UniProt Q15796; also known as hMAD-2, hSMAD2, JV18-1, MAD homolog 2, Mad-related protein 2, Mothers against DPP homolog 2, SMAD 2, SMAD family member 2, Smad2) is encoded by the SMAD2 (also known as MADH2, MADR2) gene (Gene ID 4087) in human. Smad2 is a receptor-regulated intracellular signal transducer and transcriptional modulator activated by transforming growth factor TGF-beta and activin type 1 receptor kinases. In response to TGF-beta signal, smad2 is recruited to the activated TGF-beta receptor through its interaction with the SMAD anchor for receptor activation (SARA) protein, where it is phosphorylated by the TGF-beta receptor. The phosphorylation results in its dissociation from SARA and association with another family member smad4. The smad2-smad4 complex then translocates into the nucleus, where it binds to target promoters and forms a transcription repressor complex with other cofactors. Smad2 can also be phosphorylated by activin type 1 receptor kinase, and thereby transduce activin-dependent signaling events.

Specificity

This antibody detects Smad2 Ser465/467 phosphorylation in TGFbeta-stimulated, but not unstimulated HepG2 cells. The corresponding target phosphoserine residues in the alternatively spliced human Smad2Deltaexon3 (Isoform Short) are pSer435 & pSer437 (UniProt Q15796-2).

Western Blotting Analysis: 2.0 µg/mL of this antibody detected Smad2 Ser465/467 phosphorylation in 10 µg lysate from TGFbeta-stimulated, but not unstimulated, HepG2 cells.
