General description

Recombinant, mouse secreted frizzled related protein 3 (sFRP-3) consisting of amino acids Ala 33-Ser323, fused at the N-terminus to a His•Tag sequence and expressed in NSO-derived mouse myeloma cells. sFRP-3 is a soluble antagonist of Wnt signaling. It prohibits interactions with Frizzled receptors by binding directly to Wnt proteins.

Biochem/physiol Actions

Proliferation Assay:

ED₅₀ = 1.6-6 µg/ml as measured by inhibition of proliferation of MC3T3-E1 mouse osteoblastic cells.

Functional ELISA:

50% optimal binding response = 150-250 ng/ml as measured by binding ro rmWnt-3a (500 ng/ml).

Warning

Toxicity: Standard Handling (A)

Physical form

Lyophilized from 0.2 µm filtered PBS, BSA.

Reconstitution

Following reconstituion refrigerate (4°C) for short term storage or aliquot and freeze (-20°C) for long-term storage. Stock solutions are stable for up to 1 month at 4°C or for up to 3 months at -20°C.
