General description

A cell-permeable chalcone derivative that preferentially increases primary and mature miR-34a levels in Huh7 and HepG2 hepatocellular carcinoma cells (HCC) expressing wild-type or mutated p53, but does not affect miR-34a expression in non-tumorigenic Hep3B cells with deleted p53. Inhibits the luciferase activity in a HCC cell-based miR34a luciferase reporter system (IC50 = 3.8 µM). Causes a significant reduction in the mRNA and protein levels of cyclin D1 and Bcl-2 in Huh7 and HepG2 cells (~ 10 µM), but not in Hep3B cells. Also shown to reduce the levels of miR-34a targets (Cdk6, FOXP, Notch 1 and SIRT) and p53 targets (p21 and PUMA) in Huh7 and HepG2 cells, but not in Hep3B cells. Inhibits the growth of multiple HCC by inducing apoptosis and reducing proliferation, but shows no toxicity on non-tumorigenic human hepatocytes. Suppresses the growth of Huh7 and HepG2 xenografts in nude mice model (~90% inhibition at 50 mg/kg).
