General description

Full length, recombinant, human histone deacetylase 8 (HDAC8) fused to at the C-terminus to a His•Tag sequence at and expressed in S. frugiperda insect cells using a baculovirus expression system. HDAC8 phosphorylated in vivo by PKA results in decreased activity, which leads to the hyperacetylation of histones H3 and H4. HDAC8 may play a role in one of the most frequent types of acute myeloid leukemia (AML). The active deacetylase is useful in the study of HDAC8 regulation and for inhibitor screening.

Unit Definition

One unit is defined as the the amount of enzyme required to deacetylate 1 picomol fluorescent peptide substrate [Arg-His-Lys(Ac)-Lys(Ac)] per min at 30°C.

In 138 mM NaCl, 25 mM Tris-HCl, 10% glycerol, 0.05% TWEEN®-20 detergent, pH 8.0.

Reconstitution

Following initial thaw, aliquot and freeze (-70°C).
