General description

The function, processing and ultimate fate of polyubiquitinylated proteins is implied to be determined by the linkage between adjacent ubiquitin molecules in the polyubiquitin chain. The post-translational modification of proteins with polyubiquitin chains occurs in a wide range of signalling pathways and is tightly regulated in order to ensure cellular homeostasis. Several ubiquitin polygenes are encoded in eukaryotic cells and undergo post-translational processing to generate the cellular source of free ubiquitin monomers. However, linear polyubiquitin chains have been shown to be assembled in vitro by an E3 ligase complex, known as the linear ubiquitin chain assembly complex (LUBAC), and ubiquitin binding domains (UBDs) with a preference for linear polyubiquitin have been identified in a number of proteins. Recent studies suggest that linear polyubiquitin modification of proteins may play an important role in cellular processes in addition to that of lysine-linked polyubiquitin protein conjugation. Aditionally, studies suggest a possible role for linear polyubiquitin modification in NF-κB pathway activation, involving linear polyubiquitinylation of NEMO.

Western Blotting Analysis: 0.14 µg/µL of Cat. No. MABS199, Anti-linear Ubiquitin, clone 1E3 detected Di-Ubiquitin (Linear) Recombinant Protein in a panel of polyubiquitin proteins.

Physical form

Format: Purified
Liquid. 50 µg of Di-Ubiquitin reconstituted in 2.5 µL DMSO and 56 µL of 0.02 M TRIS, 0.25 M NaCl, pH 7.5 to a final concentration of 50 µM. General used concentrations vary from 10 – 50 μM. Buffer exchange using 3 kDa spin filters (or dialysis membrane) can be used to remove the DMSO if desired.

Storage and Stability

Store reconstituted solution at −80°C. Please avoid multiple freeze/thaw cycles.
