Gene Information

human ... ALKBH3(221120)

General description

ABH3 is a member of the Fe(II)/2OG-dependent dioxygenase superfamily, which couples substrate oxidation by converting 2-oxoglutarate (2OG) into succinate and CO2. In humans, ABH3 plays a critical role in alkylated DNA/RNA damage repair and in cancer therapy. While ABH3 may repair alkylated RNA, it demonstrates greater preference for the repair of single- over double-stranded alkylated DNA through oxidative demethylation of 1-methyladenine and 3-methylcytosine.

Specificity

Other homologies Orangutan 98% and Rhesus Monkey 97% sequence homology.
This antibody recognizes the N-terminus of ALKBH3.

Immunogen

Transfer RNA is heavily modified and plays a central role in protein synthesis and cellular functions. Here we demonstrate that ALKBH3 is a 1-methyladenosine (m1A) and 3-methylcytidine (m3C) demethylase of tRNA. ALKBH3 can promote cancer cell proliferation, migration and invasion.

The AlkB repair enzymes, including Escherichia coli AlkB and two human homologues, ALKBH2 and ALKBH3, are iron(II)- and 2-oxoglutarate-dependent dioxygenases that efficiently repair N(1)-methyladenine and N(3)-methylcytosine methylated DNA damages. The mammalian AlkB homolog (ALKBH) family of proteins possess a 2-oxoglutarate- and Fe(II)-dependent oxygenase domain. A similar domain in the Escherichia coli AlkB protein catalyzes the oxidative demethylation of 1-methyladenine (1-meA) and 3-methylcytosine (3-meC) in both DNA and RNA. AlkB

ALKBH3, a demethylase responsible for demethylating N1-methyladenosine (m1A) in mRNA and N1-methyldeoxyadenosine in single-stranded DNA, plays an important role in DNA repair and cancer cell proliferation. However, its function in hematopoietic stem cells (HSCs) is unknown. In this study, we
