General description

PRKCA-binding protein (UniProt Q62083; also known as Protein interacting with C kinase 1, Protein kinase C-alpha-binding protein) is encoded by the Pick1 (also known as Prkcabp) gene (Gene ID 18693) in murine species. PICK1 is a scaffolding protein expressed in many tissues, where it regulates trafficking of multiple membrane proteins. The PDZ (PSD-95/Dlg/ZO-1/DHR (Dlg homologous region) domain (a.a. 22-105) mediates PICK1 interaction with proteins containing PDZ-binding motifs, such as PKC and acid sensing ion channels (ASICs), while the lipid-binding AH (arfaptin homology)/BAR (Bin/amphiphysin/Rvs) domain (a.a. 144-357) mediates PICK1 homo-dimerization and membrane localization. Pick1 gene-disruption results in downregulated ASICs current and calcium transients during process of ischemia-reperfusion and protects against primary murine cortical neurons against injury caused by OGD-Rep (oxygen and glucose deprivation followed by reperfusion). PICK1 is also shown to act as a negatively regulator against ASIC1 phosphorylation/activation by PKA in rat pulmonary arterial smooth muscle cells, where PICK1 facilitates calcineurin-mediated ASIC1 dephosphorylation.

Specificity

This rabbit polyclonal antibody targets the C-terminal region of mouse and rat PICK1, as well as human PICK1 spliced isoform 1, but not isoform 2, reported by UniProt (Q62083, Q9EP80, Q9NRD5).

Anti-PICK1, Cat. No. 07-293-I, is a highly specific rabbit polyclonal antibody that targets PRKCA-binding protein and has been tested in Immunohistochemistry (Paraffin) and Western Blotting.
Immunohistochemistry Analysis: A 1:1,000 dilution from a representative lot detected PICK1 in human cerebellum, kidney, and pancreas tissue sections.

Evaluated by Western Blotting in PC12 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PICK1 in 10 µg of PC12 rat pheochromocytoma cell lysate.

Target description

~55 kDa observed. 46.60 kDa (human/mouse) and 46.61 kDa (rat) calculated. The larger-than-calculated band size is consistent with that reported in the literature (Xia, J., et al. (1999). Neuron. 22(1):179 187).
