Gene Information

General description

Insulin-like growth factor I (UniProt P05019; also known as IGF-I, Somatomedin-C) is encoded by the IGF1 (also known as IGFI, MGF) gene (Gene ID 3479) in human. Human insulin-like growth factor-I (IGF-I) is a single chain 70 amino acid polypeptide that shares structural homology with insulin and human IGF-II. IGF-I plays a critical role in growth, differentiation and development via autocrine, paracrine and endocrine mechanisms. It is a mitogen and also has effects on cell migration and differentiation. IGF-I is a regulator of metabolism and is also implicated in the pathophysiology of cancer, diabetes and obesity. Upon binding the tetrameric (alpha:beta)2 IGF-I receptor, the receptor alpha chains induce the tyrosine autophosphorylation of the beta chains, which in turn trigger cell type-specific downstream signaling cascade of events.

Specificity

IGF-I (Russel, W.E., (1984). Proc. Natl. Acad. Sci. U. S. A. 81(8):2389-2392). 40% cross reactivity to IGF-II (Van Wyk, J.J., and Bruton, E.T. (1997). Endocrinology. 138(10):4521-4522).

Immunogen

Highly purified human plasma insulin-like growth factor I (IGF-I; Somatomedin C; Sm-C) conjugated to mouse albumin (Van Wyk, J.J., and Bruton, E.T. (1997). Endocrinology. 138(10):4521-4522). Action of somatomedins on cell growth: Effect of selective neutralisation of somatomedin C (IGF-I) with a monoclonal antibody. In: Raiti, S., Tolman, and R.A. (eds.), Human Growth Hormone. pp. 585–599. Plenum Publishing Corp., New York, NY).

Application

Anti-IGF-I Antibody, clone Sm1.2, Alexa Fluor 488 Conjugate is an antibody against IGF-I for use in Immunocytochemistry.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected IGF-I in HeLa and NIH/3T3 cells.
The unconjugated antibody (Cat. No. 05-172) is shown to be suitable also for Western blotting, immunohistochemistry, immunoprecipitation, and neutralization applications.
