General description

Phosphatidic acid (PA) is a simple cellular glycerophospholipid with a negative charge and constitutes 1-4% of membrane lipids.[1] It possesses a phosphomonoester polar head group and two long hydrophobic acyl chains.[1] 7-nitro-2-1,3-benzoxadiazol-4-yl (NBD) PA is a fluorescently labelled lipid with 7-nitrobenz-2-oxa-1,3-diazol-4-yl (NBD) at the sn-2 fatty acid of PA.[2]

Application

16:0-12:0 NBD PA or 1-palmitoyl-2-{12-[(7-nitro-2-1,3-benzoxadiazol-4-yl)amino]dodecanoyl}-sn-glycero-3-phosphate (ammonium salt) may be used:
  • in the preparation of phosphatidic acid (PA) small unilamellar vesicles (SUVs) to determine mammalian target of rapamycin complex 1 (mTORC1) lysosomal translocation[3]
  • to incubate cells for the imaging of lyso phosphatidylcholine (PC) in P. falciparum live cells[4]
  • as a component of separation buffer to determine the separation of phosphoinositides and sphingolipids in the microchip devices[5]
  • to measure the cytidine diphosphate diacylglycerol (CDP-DAG) synthase activity[6]

Biochem/physiol Actions

Phosphatidic acid (PA) is implicated in many membrane fusion and membrane fission events, such as fission of Golgi membranes.[1] It functions as a signaling molecule responsible for various cellular processes including membrane trafficking and sperm hyper-activation.[7]
