General description

Fluorescent Transfection Reagent has the cationic lipid 1,2-dioleoyloxypropyl)-N,N,N-trimethylammonium chloride (DOTAP) and the neutral lipid DOPE (1,2-dioleoyl-sn-glycero-3-phosphoethanolamine in the ratio 1:1 w/w along with 0.1 wt% N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)dioleoyl (NBD) DOPE. NBD-DOPE is a fluorescence label. NBD is attached to headgroup of DOPE.

Fluorescent Transfection Reagent has been used to visualize the localization of liposome oligodeoxynucleotides (ODN) complex in lung alveolar cells.

Biochem/physiol Actions

Fluorescent Transfection Reagent is useful in understanding fibroplex membrane structure and interaction of fibroin particles with cationic lipids. N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)dioleoyl (NBD) is used in model membranes studies and is useful in visualizing membrane domains, at 460/535 nm (excitation/emission) wavelengths. The DOTAP:DOPE with 0.1% NBD-DOPE liposome (LipNBD) reconstituted with transferrin called the transferrin liposome-nitrobenzoxadiazole (Tf-LipNBD) is useful as a probe. It provides improved image contrast in in vitro magnetic resonance imaging (MRI) of tumor cells.
