The procedure involves mechanical cell wall and cell membrane breakage, removal of cell debris and leaf tissue by filtration, collection of total cell chloroplast fraction by centrifugation, and separation of intact chloroplasts from broken chloroplasts using a Percoll® layer or gradient.
