Specificity

Clone 1G18 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Interleukin-6 (IL-6). It targets an epitope within 23 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the internal region of mouse IL-6.

Application

Quality Control Testing

Evaluated by Western Blotting with recombinant mouse IL-6.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected recombinant mouse IL-6.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound murine IL-6 peptide with a KD of 1.6 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected IL-6 in mouse spleen and human tonsil tissue sections.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected IL-6 in one million RAW264.7 cells.

Target description

Interleukin-6 (UniProt: P08505; also known as IL-6, B-cell hybridoma growth factor, Interleukin HP-1) is encoded by the IL6 (also known as IL-6) gene (Gene ID: 16193) in murine species. IL-6 is an immunoregulatory secreted cytokine that is synthesized with a signal peptide (aa 1-24), which is subsequently cleaved off to produce the mature form (aa 25-211). It is expressed by dendritic cells, macrophages, and activated follicular B cells. It is also abundantly expressed in the central nervous system, particularly in the hypothalamic region. It is a potent inducer of the acute phase response and rapid IL-6 production contributes to host defense during infection and tissue injury. In the innate immune response, it is synthesized by myeloid cells upon recognition of pathogens through toll-like receptors (TLRs) at the site of infection. In the adaptive immune response, it is required for the differentiation of B cells into immunoglobulin-secreting cells. It is reported to plays a major role in the differentiation of CD4+ T cell subsets and is an essential factor for the development of T follicular helper (Tfh) cells. IL-6 also acts as an essential factor in bone homeostasis and acts directly or indirectly on blood vessels by induction of VEGF that results in increased angiogenesis activity and vascular permeability. Its actions are mediated by binding to IL-6 receptor (IL-6R), and the ligand-receptor complex associates to the signaling subunit IL6ST/gp130. The interaction with the membrane-bound IL6R and IL6ST is reported to stimulate the classic signaling, whereas the binding of IL-6 and soluble IL6R to IL6ST stimulates trans-signaling where synergistically with IL-1β and TNF it can induce the production of VEGF and stimulate angiogenesis. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
