General description

Research area: Cancer

FGFR3 (fibroblast growth factor receptor 3) is a receptor tyrosine kinase (RTK) and belongs to the FGFR subfamily. This subfamily includes four members namely, FGFR1, FGFR2, FGFR3, and FGFR4. It contains a ligand-binding region in its exoplasmic domain, a single transmembrane region, and a tyrosine kinase domain in its intracellular domain.FGFR3 has a strong affinity for canonical FGF ligands.

Biochem/physiol Actions

FGFR3 (fibroblast growth factor receptor 3) plays an essential role in the signaling pathways during embryogenesis and angiogenesis. This gene is either mutated or up-regulated in multiple myeloma, cervical carcinoma and nonmuscle-invasive urothelial carcinoma (UC) of the bladder.Apart from this, certain mutations in FGFR3 are also known to be linked with skeletal disorders such as various forms of dwarfism. FGFR3 also plays hepatoprotective function during liver damage.

Fibroblast growth factor receptors (FGFRs) exist as a gene family of 4 membrane bound receptor tyrosine kinases (FGFR1-4) that mediate signals of at least 22 fibroblast growth factors (FGF1-22). FGFs/FGFRs play important roles in multiple biological processes, including mesoderm induction

FGFR3 is a receptor tyrosine kinase (RTK) of the FGF receptor family, known to have a negative regulatory effect on long bone growth. Fgfr3 knockout mice display longer bones and, accordingly, most germline-activating mutations in man are associated with dwarfism.

Activation of the protein tyrosine kinase receptors requires the coupling of ligand binding to a change in both the proximity and orientation of the single transmembrane (TM) helices of receptor monomers to allow transphosphorylation of the receptor kinase domain. We
