Target description

Macrophage colony-stimulating factor 1 (UniProt: P07141; also known as CSF-1, MCSF) is encoded by the Csf1 (also known as Csfm) gene (Gene ID: 12977) in murine species. CSF-1 is a widely expressed, single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-32), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 33-492), a transmembrane domain (aa 493-515), and a cytoplasmic domain (aa 516-552). Following its synthesis, it can be cleaved into a 43 kDa subunit CSF1 and a processed secreted form. CSF-1 is a cytokine that plays an essential role in the regulation of survival, proliferation and differentiation of hematopoietic precursor cells and promotes the release of pro-inflammatory chemokines, and thereby plays an important role in innate immunity and in inflammatory processes. It also plays an important role in the regulation of osteoclast proliferation and differentiation, the regulation of bone resorption, and is required for normal bone development. Its actions are mediated by a high affinity CSF1-receptor. Clone 5A1 is shown to bind and inhibit the biological functions of CSF-1. It is reported to inhibit colony formation of tissue mononuclear phagocyte colony-forming cells as well as the committed bone marrow stem cells for both granulocytes and monocytes. The extent of colony inhibition by this clone is dependent on the tissue origin of colony-forming cells. Two isoforms of CSF-1 have been described that are produced by alternative splicing. Purified rat monoclonal antibody IgG1 in PBS without azide.

Reconstitution

0.5 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
