General description

MUG EC broth is a selective and differential liquid culture mediumspecifically formulated for the rapid detection of Escherichia coli inmicrobiological samples. The medium is based on EC broth components—caseinenzymatic hydrolysate as the source of essential nutrients, lactose as afermentable carbohydrate, bile salts, buffer salts, and sodium chloride tomaintain osmotic equilibrium—and is supplemented with the fluorogenic substrate4‑methylumbelliferyl β‑D‑glucuronide (MUG). MUGis hydrolyzed by the E. coli enzyme β‑glucuronidase to yield the fluorescent product 4‑methylumbelliferone, which can be observed under long‑wave UV light, enabling rapid and specific detection of E. coli. Caseinenzymic hydrolysate provides essential nutrients. Lactose is a fermentablecarbohydrate. Sodium chloride maintains the osmotic equilibrium. The medium’sstrong buffering system controls pH during fermentative activity, while bilesalts inhibit gram‑positive bacteria,particularly Bacillus species and fecal streptococci, enhancingselectivity for E. coli.

Application

MUG EC broth is used as a selective and differential medium for thedetection and confirmation of Escherichia coli in water, food, andenvironmental samples, often within coliform testing schemes. Bile salts andincubation at elevated temperature (e.g., ~44.5 °C as per standard EC methods)inhibit many non‑coliform bacteria whileallowing growth of coliforms, and the MUG reagent enables differentiation of E.coli from other coliforms by fluorescence. This makes the broth suitablefor regulatory and quality-control analyses where specific enumeration orconfirmation of E. coli is required, including drinking water, recreationalwater, wastewater, and various food matrices.

Preparation Note

Suspend 37 g of MUG EC Broth in 1000 ml of distilled water. Dispense in tubes containing inverted Durham′s tubes. Sterilize by autoclaving at 15 lbs. pressure (121°C) for 12-15 minutes.
