Spécificité

Clone 2C10 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects myosin light chain 2 phosphorylated on serine 19.

Immunogène

KLH-conjugated linear peptide corresponding to 13 amino acids surrounding phosphoserine 19 from the N-terminal region of human Myosin Light Chain 2 (MLC2).

Application

Quality Control Testing

Evaluated by Western Blotting in lysates from C2C12 cells.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected phospho-Myosin light chain 2 (Ser19) in lysate from C2C12 cells.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected phospho-Myosin LC 2 (Ser19) in C6 and HEK293 cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected phospho-Myosin LC 2 (Ser19) in human uterus tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound phospho-Myosin Light Chain 2 (Ser-19) with a KD of 6.0 x 10-6 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Description de la cible

Myosin regulatory light polypeptide 9 (UniProt: P24844; also known as 20 kDa myosin light chain, LC20, MLC-2C, Myosin RLC, Myosin regulatory light chain 2 smooth muscle isoform, Myosin regulatory light chain 9, Myosin regulatory light chain MRLC1) is encoded by the MLY9 (also known as MLC2, MRLC1, MYRL2) gene (Gene ID: 10398) in human. Myosins are highly conserved, ubiquitously expressed proteins that form regular bipolar thick filaments that together with actin thin filaments constitute the fundamental contractile unit of skeletal and cardiac muscle. Along with actin they serve as motor molecules with ATPase activity that is essential for muscle contraction. Muscle myosin is a hexameric protein that consists of 2 heavy chain subunits (MHC), 2 light chain subunits (MLC) and 2 regulatory light chain subunits (MLC-2). MLC-2C plays an important role in regulation of both smooth muscle and non-muscle cell contractile activity via its phosphorylation. It is phosphorylated at threonine 18 and serine 19 by myosin light chain kinase (MLCK) in a Ca2+/CaM-dependent manner. Rho kinase (ROCK) is also known to phosphorylates MLC-2C at serine 19. Phosphorylation increases the actin-activated myosin ATPase activity and thereby regulates the contractile activity. It is required to generate the driving force in the migration of the cells. Phosphorylation is also required for myotube formation. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
