General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

Application

Quality Control Testing

Evaluated by Flow Cytometry in PMA/Ionomycin-stimulated mouse splenocytes.

Flow Cytometry Analysis: One million BALC/c mouse splenocytes were unstimulated or stimulated with 50 ng/mL of Phorbol 12-myristate 13-acetate (PMA) and 1 μg/mL of ionomycin for 6 hours, and 3 μg/mL Brefeldin A and 2 μM of monensin were added 3 hours before harvesting cells. Cells were stained with APC Anti-Mouse CD3e, fixed, permeabilized and subsequently stained with 5 μL of a 1:1 mixture of Cat. No. CWA-1087, Anti-Mouse IFN- (XMG1.2) ColorWheel® Dye-Ready mAb and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye.

Target description

Interferon - (UniProt: P01580; also known as IFN- ) is encoded by the Ifng gene (Gene ID: 15978) in murine species. IFN- is a pleiotropic molecule with antiproliferative, pro-apoptotic, and antitumor properties. It is a type II homodimeric interferon that is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to produce the mature form. It is secreted predominantly by activated lymphocytes such as CD4 T helper type 1, CD8 cytotoxic T cells, T cells, and natural killer cells, and antigen-presenting cells and plays crucial roles in antimicrobial, antiviral, and antitumor responses by activating effector immune cells and enhancing antigen presentation. Its expression is induced by mitogens and cytokines. IFN- signaling coordinates several biological responses involved in host defense and immune surveillance, establishment of adaptive immunity, and the regulation of inflammation. IFN- is reported to activate macrophages toward a pro-inflammatory profile increasing their phagocytic ability and enhancing their microbial killing activity. It is also shown to upregulate cell surface MHC class II on antigen presenting cells, thus promoting peptide-specific activation of CD4+ T cells. Its diverse biological activities are initiated by IFN- -mediated aggregation of at least two different cell surface receptors: IFN- R1 and IFN- R2. It primarily signals through the JAK-STAT pathway after interaction with IFN- R1 to affect gene regulation. Upon IFN- binding, intracellular domain of IFN- R1 opens out to allow association of downstream signaling components JAK2, JAK1 and STAT1, leading to STAT1 activation, nuclear translocation and transcription of IFN- -regulated genes. Mutations in Ifng gene are known to cause aplastic anemia that is characterized by failure of bone marrow to produce adequate number of peripheral blood elements leading to peripheral pancytopenia and hypoplasia.

Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitution

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
