For immunocytochemistry, colcemid-arrested HeLa cells were fixed with 1% paraformaldehyde. Also detects histone H3 in Tetrahymena micronuclei. The antibody has been reported to detect mitotic cells in Xenopus, Caenorhabditis elegans, and Drosophila embryos. Does not cross-react with unphosphorylated histone H3. This antibody has also been reported to work for immunohistochemistry. Variables associated with assay conditions will dictate the optimal working dilution.
