Beta-Pix (ARHGEF7, βPix, COOL) is a Rho guanine nucleotide exchange factor (GEF). Beta-Pix is a p21-activated protein kinase (PAK)-interacting exchange factor (Pix, COOL, p85SPR) that has been identified as a putative guanine nucleotide exchange factor (GEF) for Rac/Cdc42. The variants of Beta-Pix isoforms exhibit different tissue distributions. Beta-Pix-a is expressed ubiquitously in all the tissues examined, and other isoforms are expressed mainly in the central nervous system. PAK, a Ser/Thr kinase, is an important mediator of Rho family activity interacts with Beta-Pix at the SH3 domain of Beta-Pix though its proline rich region. This interaction is thought to affect the GTP hydrolysis rate of Rac and Cdc42.

Cell motility is a tightly regulated phenomenon that supports the accurate formation of organ structure during development and homeostasis, including wound healing and inflammation. Meanwhile, cancer cells exhibit dysregulated motility, which causes spreading and invasion. The Dbl family RhoGEF ARHGEF7/β-PIX
