A sensitive assay is described for quantitating dolichyl phosphate (Dol-P), the polyprenyl phospholipid which participates in N-linked glycosylation. The assay is based on a novel reaction of alkyl phosphates with phenyl chloroformate in which a monosubstituted mixed anhydride of phosphoric
