Specificity

Clone 2H18 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Tumor susceptibility gene 101 protein (TSG101). It targets an epitope within 19 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the C-terminal half of human Tumor susceptibility gene 101 protein (TSG101).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected TSG101 in HeLa cell lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected TSG101 in MCF-7, NIH3T3, and Mouse brain tissue lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TSG101 in MCF-7 cell line.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TSG101 in human colon tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound TSG101 with a KD of 7.7 x 10-8 in an affinity binding assay.

Anti-TSG101, clone 2H18 ZooMAb®, Cat. No. ZRB1244, is a recombinant Rabbit monoclonal antibody that specifically targets TSG101 and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Tumor susceptibility gene 101 protein (UniProt: Q99816; also known as ESCRT-I complex subunit TSG101) is encoded by the TSG101 (Gene ID: 7251) in human. TSG101 is a ubiquitously expressed protein that is mainly localized in the cytoplasm. However, when active it is membrane associated and is inactive when in soluble form. It is also found in the nucleus during the late S phase and co-localizes with spindle apparatus during mitotic cell division. Human TSG101 contains four known structural domains: an N-terminal UEV domain, followed by a proline-rich region, a coiled coil region, and a C-terminal -helical/steadiness box domain. TSG101 mediates a variety of intracellular processes, including cell growth and differentiation, but its major role in in endosomal trafficking of cargo proteins. It is a component of the ESCRT-I complex that associates with VPS28 and VPS37 protein. It binds to ubiquitinated cargo proteins and is required for the sorting of endocytic ubiquitinated cargos into multivesicular bodies (MVBs). It is also involved in the extracellular release of micro vesicles that differ from the exosomes. TSG101 gene is considered as a housekeeping gene and its knockout is reported to cause embryonic lethality. Loss of TSG101 is reported to trigger an accumulation of p53 and cell cycle arrest leading to cell death. Although it is considered as an tumor suppressor, its elevated levels of are observed in some epithelial cancer cells. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
